چکیده

A rapid method for nearly quantitative isolation of human serum albumin is described. The present technic (1) is a modification of the Schwert method (2). In the present method 4 ml. of serum is precipitated with trichloroacetic acid in a final concentration of 5% (w/v). The precipitate is washed with TCA, and after aqueous suspension, ethanol is added to a final concentration of 80%. The undissolved globulin is washed with ethanol and the solubilized albumin used for quantification, characterization, and isotope counting. Recovery of added human serum albumin was approximately 98%; recovery of added 1311 albumin was 90%. Cellulose acetate electrophoresis showed a single band, and agar diffusion produced one precipitate line. Immunoelectrophoresis with anti-human serum revealed one or two very faint arcs in the a-, or aand ?-globulin regions, in addition to the major arc of albumin. The method has shown excellent reproducibility in replicate analyses of normal and pathologic serums.

Isolation and purification of albumin from human plasma by direct and combined approach of ion-exchange chromatography and comparison of the final products quality obtained by both methods

Background: Due to multiple roles of albumin in the body, injection of its medicinal product as one of the therapeutic or management strategies under conditions such as severe bleeding, burns, liver failure, and neonatal hemolytic diseases is on the physicians' agenda. Considering that albumin is the most abundant plasma protein, designing an appropriate method to purify it is highly important....

استفاده از اتانول و دمای پایین در جداسازی آلبومین از پلاسمای انسانی

Background and Objective: Human plasma is a suitable source for different proteins and enzymes with a variety of therapeutic effects. Examples include coagulation factors VIII and IX, immunoglobulin, and albumin, which consists of a 585 amino-acid chain, weighing 66.5 kD. Albumin molecule also has 17 di-sulfide bridges which form a stable spherical protein structure, which it cause of 80% of on...

تهیه داروی بیولوژیکی آلبومین از پلاسمای انسانی با استفاده از فیلتراسیون فیبری

Background: In recent years, consumption of whole-blood for the treatment of patients has decreased but use of biological plasma-derived medicines such as albumin, immunoglobulin and coagulation factors have increased instead. Paying attention to albumin molecular structure is important for its isolation from human plasma. Albumin is a single-chain protein consisting of about 585 amino acids an...

Characteristics of Potential Protein Biomarkers Extracted with 10% TCA from Blood Serum of Non-Hodgkin’s Lymphoma and Multiple Myeloma Patients

Blood serum has been extensively explored as a source of the bio-markers [1, 2].  For concentration of minor protein(s) and depletion of abundant blood serum proteins a 2,2,2-trichloroacetic acid (TCA) precipitation procedure is frequently applied [3]. However, a significant amount of proteins may be present in the TCA extracts, and these proteins are often not studied.Recently, we have sh...
